Efficient characterization of retro-, lenti-, and foamyvector-transduced cell populations by high-accuracy insertion site sequencing

The identification of unknown genomic flanking DNA sequences can be used for the molecular monitoring of retro-, lenti- and foamyviral integration, transgenes in early embryogenesis, insertional mutagenesis, cell fate, and stem cell plasticity. Most existing methods reflect shortcomings in sensitivity and or specificity, thus limiting genomic sequencing of unknown flanking DNA to clonal preparatio

Superantigen- and TLR-dependent activation of tonsillar B cells after receptor-mediated endocytosis.

Classical B lymphocyte activation is dependent on BCR cross-linking in combination with physical interaction with Th cells. Other B cell molecules that contribute to the activation are complement, cytokine, and TLRs recognizing specific pathogen-associated molecular patterns. Moraxella (Branhamella) catarrhalis is a common Gram-negative respiratory pathogen that induces proliferation in human IgD-

The spatiotemporal distribution of human pathogens in ancient Eurasia

Infectious diseases have had devastating effects on human populations throughout history, but important questions about their origins and past dynamics remain1. To create an archaeogenetic-based spatiotemporal map of human pathogens, we screened shotgun-sequencing data from 1,313 ancient humans covering 37,000 years of Eurasian history. We demonstrate the widespread presence of ancient bacterial,

Mitoxantrone alters CD24/Siglec-10 expression in malignant brain tumor models

Medulloblastoma and glioblastoma are the most common malignant primary brain tumors in children and adults, respectively. Tumor-associated macrophages and microglia are key non-cancerous cell types in these tumors. These cells interact with CD24, a so called “don’t eat me signal” expressed on tumor cells, through Siglec-10, a receptor that contributes to immune evasion by promoting an immunosuppre

ClusterVAP : study protocol for multicentre proteomic endotyping of ventilator-associated pneumonia

INTRODUCTION: Ventilator-associated pneumonia (VAP) is the most frequent healthcare-associated infection in intensive care units and is associated with high morbidity and mortality. Current diagnostic criteria lack specificity, leading to misclassification and unnecessary antibiotic use. Identifying patient subgroups with a common pathophysiological basis (pneumoclusters) may distinguish true VAP

Exome analysis links kidney malformations to developmental disorders and reveals causal genes

Congenital anomalies of the kidneys and urinary tract (CAKUT) are developmental disorders that commonly cause pediatric chronic kidney disease and mortality. We examine here rare coding variants in 248 CAKUT trios and 1742 singleton CAKUT cases and compare them to 22,258 controls. Diagnostic and candidate diagnostic variants are detected in 14.1% of cases. We find a significant enrichment of rare

Event driven design methodology for hardware DSP design

Today's hardware DSP designs, with a composition of hardware in datapath/controller pairs, are becoming increasingly complex to design. It will be argued that hardware DSP architectures can be significantly simplified by distributing the control to the individual pipeline stages and by switching to an event based design methodology. This paper will describe a design methodology for event based DSP

The GALAH survey : Chemical homogeneity of the Orion complex

Due to its proximity, the Orion star forming region is often used as a proxy to study processes related to star formation and to observe young stars in the environment they were born in. With the release of Gaia DR2, the distance measurements to the Orion complex are now good enough that the 3D structure of the complex can be explored. Here we test the hypothesis that, due to non-trivial structure

The human gut microbiome in early-onset type 1 diabetes from the TEDDY study

Type 1 diabetes (T1D) is an autoimmune disease that targets pancreatic islet beta cells and incorporates genetic and environmental factors1, including complex genetic elements2, patient exposures3 and the gut microbiome4. Viral infections5 and broader gut dysbioses6 have been identified as potential causes or contributing factors; however, human studies have not yet identified microbial compositio

Hand eczema in healthcare workers : a questionnaire survey of risk factors and work-related exposures

Recent data on the prevalence and causes of hand eczema among healthcare workers in Sweden are lacking. Multidrug-resistant bacteria have necessitated improved hand hygiene and preventive measures. This has led to an increase in the use of disposable rubber gloves and hand disinfectants, which might influence the risk of hand eczema. Our aims were to identify healthcare workers with hand eczema; t

Automated pathway and reaction prediction facilitates in silico identification of unknown metabolites in human cohort studies

Identification of metabolites in non-targeted metabolomics continues to be a bottleneck in metabolomics studies in large human cohorts. Unidentified metabolites frequently emerge in the results of association studies linking metabolite levels to, for example, clinical phenotypes. For further analyses these unknown metabolites must be identified. Current approaches utilize chemical information, suc

European EADV network on assessment of severity and burden of Pruritus (PruNet): first meeting on outcome tools.

Chronic pruritus is a frequently occurring symptom of various dermatoses that causes a high burden and impaired quality of life. An effective anti pruritic therapy is important for the patient, but its effectiveness is difficult to evaluate. Diverse methods and interpretations of pruritic metrics are utilized in clinical trials and the daily clinical practice in different countries, resulting in d

Little Doubt That CBT Works for Pediatric OCD

We write with great concern in response to the recent systematic review and meta-analysis of cognitive-behavioral therapy (CBT) in pediatric obsessive-compulsive disorder (OCD) by Uhre et al.1 Although the authors’ results consistently support the clinical efficacy of CBT for pediatric OCD, we expect that, much like ourselves, readers will be confused by the discordant and inappropriate conclusion

LUCAS versus manual chest compression during ambulance transport : A hemodynamic study in a porcine model of cardiac arrest

Background—Mechanical chest compression (CC) is currently suggested to deliver sustained high-quality CC in a moving ambulance. This study compared the hemodynamic support provided by a mechanical piston device or manual CC during ambulance transport in a porcine model of cardiopulmonary resuscitation. Methods and Results—In a simulated urban ambulance transport, 16 pigs in cardiac arrest were ra

Evidence for the formation of comet 67p/churyumov-gerasimenko through gravitational collapse of a bound clump of pebbles

The processes that led to the formation of the planetary bodies in the Solar system are still not fully understood. Using the results obtained with the comprehensive suite of instruments onboard the European Space Agency's Rosetta mission, we present evidence that comet 67P/Churyumov-Gerasimenko likely formed through the gentle gravitational collapse of a bound clump of mm-sized dust aggregates ('

Health, cattle and ploughs : Bioarchaeological consequences of the Secondary Products Revolution in southern Sweden, 2300-1100 BCE

In this thesis diet and health of people who lived in southern Sweden 2300-1100 BCE is studied. The study is based on bioarchaeological analyses of human remains from 46 localities in the areas of Uppland, Närke, Östergötland, Västergötland, and Scania. The studies are based on skeletal remains from a minimum number of 310 individuals that have been analysed both osteologically and biochemically.
